 Ex-Pure is a monolithic silica column for antibody purification.

 Three types are available for the Ex-Pure, and those are divided into two types,
Protein A and
Protein G
, respectively.

Spin-type

 Antibody purification in a short time becomes possible by immobilizing the Protein A and Protein G
on theplate-shaped monolithic silica surface.

 Purification and concentration of target IgG antibody, and measurement of IgG antibody
concentrations in the culture solution can be performed easily in two minutes.

 *The high-density chemical bonding technology into a modified protein G and the silica surface
was invented and developed by the National Institute of Advanced Industrial Science and Technology
Institute of Technology.

 Using this technique, monolithic silica column for antibody purification (Ex-Pure) has been
developed and manufactured at Kyoto Monotech.

Performance of spin column
Protein A(ProA) Protein G(ProG)
 Human IgG up to 0.4 mg can be purified at a
recovery rate of
90% or more by using a spin
column ligand with the Ex-Pure ProA.
 In addition, amount of protein up to 0.3 mg can be almost
quantitatively measured in each step.
 A spin column ligand with the Ex-Pure ProG also can purify and recover antibody as easily as the
Ex-Pure ProA, and the amount of protein in liquid passage can be quantitatively measure in each
step.
 Furthermore,
IgG3 type of antibody, which
cannot bind to Ex-Pure ProA, can also be purified
and recovered.



Evaluation of antibody recovery accuracy (variation) of the spin column
 UV method Protein A(ProA) Protein G(ProG)
0.65±0.027mg/column 0.504±0.056mg/column
 BCA method 0.641±0.079mg/column
 A spin column was selected at random, the human IgG of 2.0 mg/mL was injected 0.4mL into it, and
washing to elution were performed.
 The antibody content in elute was measured using the UV method (A280) and the BCA method.
*For Ex-Pure ProG, only the UV method was used.


Antibody purification evaluation from the culture fluid by SDS-PAGE electrophoresis
 It shows the result of SDS-PAGE when antibody was actually purified from the culture using the
Ex-Pure.
 As you can find, contaminants have been removed at the lane 3 (flow-through solution) and at the
lane 4 (cleaning solution).


96Well Plate-typ


 Liquid passage approx.1mL/min. is possible under reduced pressure of 0.2 atm.

 Recovery rate equivalent to with the spin column type can be obtained.

 12mg of human IgG antibody will be recovered by ProA.
